Sažetak
Liver regeneration is an essential component of the reparative process following injury, accompanied by increased reactive oxygen species production and oxidative damage, that can be prevented by reinforcement of the endogenous defense mechanism through the intake of dietary antioxidants, such are polyphenols. Hence, the effect of the olive oil polyphenolic extract (PFE) on the serum antioxidant levels and on the hepatic lipid peroxidation (LP) following 1/3 partial hepatectomy (pHx) was studied in mice, along with implications of these effects on the liver regeneration. Additionally, in vitro antioxidant capacity in terms of DPPH radical scavenging ability was assesed. For that purpose, C57/BL6 mice were assigned in four groups: PFE-mice treated with polyphenolic extract, pHx-mice subjected to 1/3 partial hepatectomy, PF+pHx-mice treated with polyphenolic extract followed by partial hepatectomy, controls-mice receiving vehicle alone.PFE was prepared by liquid-liquid extraction and intraperitoneally injected on 7 successive days in a dose of 50 mg/kg bwt. Mice were sacrificed 24h after pHx. LP was established by measuring malondialdehyde (MDA) in liver homogenates using thiobarbituric acid reaction. General levels of antioxidants were evaluated by measurements of the serum total antioxidant capacity (TAC). A calculated relative liver weight was used to assess the liver regeneration ratio. In vitro data elicited that PFE was 1.5 more effective radical scavenger than α-tocopherol. In the animal model and with respect to controls, pHx induced marked increase in the liver MDA level, as well as an active consumption of serum antioxidants. Although the level of MDA in PFE+pHx group was higher than in the control group, treatment with PFE prior to pHx reinforced TAC depletion and significantly reduced liver MDA level when compared to pHx animals. Furthermore, the relative liver weight in the PFE+pHx group was significantly higher than in the pHx group. Consequently, pre-treatment with PFE has beneficial effect on the liver regeneration, presumably due to its antioxidant properties, as indicated by attenuated LP level and enhancement of the serum antioxidant status.
