Sažetak
The active role of adipose tissue as an endocrine organ was first recognised in adipocytokine secretion. Beside their influence on metabolic balance, they also directly participate in the development of the state of chronic inflammation in obesity. Adiponectin, especially high molecular weight - HMW adiponectin is prominent among adipocytokines because of his protective metabolic and antiinflamatory actions. Immune cells like macrophages in adipose tissue, but also their antecedents, monocytes in circulation, also contribute to the state of chronic inflammation in obesity, The aim of this study was to evaluate whether there is a relation between serum HMW adiponectin level and other metabolic and inflammatory factors with monocyte activation. The study was performed in postmenopausal women with different degree of obesity. Monocytes from peripheral blood were analyzed by quantitative flow cytometry in order to measure expression of adhesion molecule CD11b, chemokine receptor CCR2 and intracellular oxidative burst with the dye dichlorofluorescin diacetate, DCFH-DA. In the study group the number of monocytes was associated with insulin and HOMA index. This correlation remained after adjusting for body mass and C-reactive protein level. The number of monocytes was also an independent predictor of insulin level in multiple regression model. The expression level of cytokine receptor CCR2 on monocytes was associated with body weight, body mass index and HOMA index. Fluorescence intensity of DCFH as a measure of oxidative burst in monocytes was correlated with anthropometric measures of obesity, fat mass but also with the expression of CCR2. The expression of integrin receptor CD11b was associated with HDL-cholesterol. There was no indication of association between HMW adiponectin and any of the analyzed markers of monocyte activation. HMW adiponectin in our sample most strongly correlated with HDL-cholesterol. These results have suggested important relations of markers of monocyte activation with measures of obesity and especially with insulin resistance. This could contribute to the understanding of the role of monocytes in metabolic disorders and warrant further studies.
