Sažetak
It has been established that after the interferon (IFN) treatrnent general morphological changes occur in cells, concomitantly with the changes in the organization of the cytoskeleton. The experiments presented here were aimed at detecting the changes in the organization of actine filaments in human embryonic fibroblasts after the treatment with homologous human alpha IFN. Cells were cultivated on small glass slides in Eagle's medium supplemented with 10% foetal calf serum (FCS) (Flow). The day before the experirnent the cells were exposed to Eagle's medium supplemented with 2% FCS. After washing the cells were treated with 500 and 1000 unit/ml of IFN having the specific activity of 104-105 units/mg of protein. This time scale was used: 15, 30, 60, 90 and 180 minutes. After the treatment the cells were washed with PBS (phosphare buffer saline, pH 7.4) and fixed with cold acetone (-20 ºC for 45 minutes. The treatment with monoclonal antiactine IgG followed and, after being washed with mouse-antirabbit IgG, the cells were conjugated with FITC. These changes have been observed by f1uorescencemicro-scopy: condensation of actine filaments and general morphological changes after 90-180 minutes of incubation.
