Sažetak
Langerhans cell (LC) has immunologic identity as antigenpresenting cell (APC) of the cutaneous immune response but LC is in toxic contact reaction involved too as target ccll for cytotoxic T-lymphocytes. In pathogenesis of irritant contact dermaritis were studied morphology, topography and frequency of LCs by histoenzymatic adenosintriphospharase (ATP-ase) method, Calcium activatcd (Ca++) ATP-ase in experimental model of irritation with 1% phenol on guinea-pig skin biopsies. Ca++ activated ATP-ase was made according to Ando et al., 1981. Morphometric measurements were made for quantification of LCs after 1, 3, 6 and 24 hours of irritation. Langerhans cells were quantified in each histological section from biopsiesof quinea pigs detected by the calcium activated ATP-ase merhod. On each 6 µm sample, the LCs were calculatcd on 10 test fields of a ocular net system B 25 in light microscope with magnification of X 640. The diametcr of the net was 0.6 mm2. The mean value of the numberof LCs per mm: for each sample was calculated. Before the experiment the nurnber of LCs was 18.1 ± 2 SE. During irritation to 1% phenol (1, 3, 6, 24 hours) and 1 month after LCs were studied and calculated. From results obtaincd it was seen 1 hour after irritation expressed linear and courinual mostly suprabasal located Ca" ATP-ase activiry in LCs and morphornetric number was 22.1 ± 3 SE. Three hours after there were still expressed LCs (20.5 ± 2 SE). Six hours after irritation LCs were continually and lincar locatcd but very expressed (26.4 ± 2 SE). Intensive stromal reaction of the dermis was seen. Rare ramify LCs were found in the epidermis 24 hours after irritation, some zone with unregulary ramify LCs distribution and expressed dendrites mostly in dermis.
