Naglic1, B. Sostaric2, Kristina Busch3 1Department of Microbiology and Infectious Diseases, Veterinary Faculty University of Zagreb, 10000 Zagreb, Heinzelova 55, Croatia; 2Croatian Veterinary Institute Zagreb, Croatia; 3PLIVA d.d., Research Institute, Zagreb, Croatia Introduction Mycoplasmoses represent the most important group of infectious diseases in intensive goat production, especially among large dairy goat farms. There are several and well recognized mycoplasmoses of goats characterized with high morbidity and mortality of adult animals and their young offspring. It should be emphasized that pathogenicity is not statically permanent to any of mycoplasmas, and that the condition of the herd affected is of crucial importance for the development, progress and the outcome of epizootic. The aim of this work is to present an outbreak of mycoplasmosis on certain dairy goat farm in Croatia inflicted with Mycoplasma capricolum subsp. capricolum. Case history The farm in question was located in Croatia and maintained over 2000 Alpino and Saanen goats imported to the country from France and Germany during last three years. There was the history of increased kid losses during previous kidding seasons as well. At the year of observation starting during the February and lasting till the late summer an epizootic characterized with high kid morbidity and mortality occurred. Totally over 1000 kids were lost during the outbreak. Extensive field and laboratory investigations were performed in order to diagnose the disease in question. Material and methods In addition to the epizootiological and clinical data collected on the farm, on the waste majority of succumbed animals autopsy was performed. Histopathological and bacteriological examinations of pertinent material were undertaken in 20% of all cases confined to autopsies. Considerable attention was paid to isolation of mycoplasmas, and for that purposes materials from different organs were inoculated and cultivated on modified Hayflick media. Results Only young kids, varying in age from 1-3 months were affected, while among goats no serious clinical disease was noted. There was obvious polyarthritis in virtually every affected animal. The articulations of extremities were swollen, warm and painful, and many of diseased animals progressively losing the ability to move were in lateral recumbence. There was a somewhat characteristic dorsoflection of head. However the appetite was preserved and animals if properly fed survived 2-3 weeks as recumbent. The intended treatment with tylosin at that stage usually was unsuccessful. Severe polyarthritis involving all extremity joints and frequently atlantoocipital junction was the most constant finding. Arthritises were characterized by heavy deposits of yellow colored fibrin intra- and periarticularly. Fibrinous pleuritis and peritonitis varying in severity were observed in more than 50% of the cases. In addition, there was an generalized enlargement of lymph nodes. In many cases there was a suppurative bronchopneumonia, and infrequently that lesion was a cause of death. Intraarticular masses microscopically were determined as fibrin strains with modest admixture of PMNs. In uncomplicated cases lungs exhibited an interstitial pneumonia characterized with type II pneumocites proliferation and diffuse alveolar walls thickening. Suppurative bronchopneumonia recognized on autopsy were readily confirmed histologically. In none of examined cases the lesions of CNS were recognized. From suppurative lung processes Pasteurella multocida, P. haemolytica, E. coli, Staphylococcus spp. and Corynebacterium spp. in decreasing order were isolated. In joint contents occasionally Actinomyces pyogenes or Escherichia coli were found. Mycoplasmas (31 isolates) were the most frequently isolated from arthritic joints, but also from lungs with interstitial pneumonia, liver and lymph nodes of necropsied kids. The isolates from all the materials except some lung samples were fast growing, large colonies forming mycoplasmas, which were dextrose-positive, arginin-negative, "film and spots"-negative, phosphatase-positive, with the ability of tetrazolium chloride reduction and sensitive to digitonine (6-7 mm). Five out of 11 lung isolates were small colonies forming slowly growing mycoplasmas, which differed from the others, being dextrose-negative, arginin-positive and phosphatase-negative. Two dextrose-positive and one arginin-positive strains were in France serologically identified as M. capricolum subsp. capricolum and M. arginini, respectively. Discussion and conclusions Epizootiological and clinical observations of epizootic with high kid mortality were indicative of mycoplasmosis. The pathoanatomical findings of fibrinous polyarthritis and frequent fibrinous pleuritis and peritonitis supported mycoplasmosis as well as histopathological finding of interstitial pneumonias. High number of bacterial pneumonias diagnosed on autopsy and confirmed bacteriologically and histopathologicaly is considered as secondary to mycoplasmosis. The diagnosis was enforced by numerous isolations of large colonies forming dextrose-fermenting mycoplasmas. Two of them being serologically identified as M. capricolum subsp. capricolum. A caprine arthritis-encephalitis lentivirus infection, a disease somewhat resembling epizootiologicaly mycoplasmosis was differentiated clearly by the fact that in our case younger animals were affected, and none of characteristic lesion of CAE was found neither serological test were indicative.
