Sažetak
Transplantable mouse methylcholanthrene induced fibrosarcoma (CMC4 tumor growing in CBA/HZgr mice), characterized by lung metastases developing shortly after local tumor cell transplantation, was used as an experimental model to investigate the problem of tumor metastases after local tumor treatment. Surgery and/or irradiation were performed on locally growing tumor of particular size. Further, heavily irradiated, viable but not dividing tumor cells, imitating the situation in treated tumor bearing organism, were injected intraperitoneally (ip.) in a parallel group of treated tumor-bearing mice. The animals were killed 35 days after tumor transplantation an the number and volume of lung metastasis were determined. Depending to the treatment performed, when tumor mass was reduced or even eliminated, the number of lung metastases and their volume were significantly lower then in control mice, but the addition of tumor mass (injection of heavily irradiated tumor cells) resulted in a significant increase in lung metastases parameters pointing to a possible role of hosts immune reaction against the tumor. Further, the release of simple molecule, as nitric oxide, from tumor mass seems to be detrimental for the survival of tumor cells and subsequently their metastases through the induction of angiogenesis and possible suppression of immune reaction. Thus, complex mechanisms could be involved if a locally growing tumor is exposed to a particular therapeutical approach.
