Sažetak
Streptomyces rimosus synthesise 19-carbon polyketide antibiotic oxytetracyline (OTC) by a 'Type II' polyketide synthase (PKS) complex. OTC biosynthetic gene-cluster occupies a contiguous 34 kb segment on the S. rimosus chromosome. One open reading frame, designated otcC, was showen to encode a hydroxylase involved in hydroxylation of C-6 of the OTC backbone. The disruption of otcC gene by the gentamicin (gmr) resistance cassette resulted in the strain producing polyketides of shorter chain length (17 C atoms). Therefore, disruption leads in 'derailment' of the ability of the OTC PKS to form a chain of the usual length. The otcC gene modified by in vitro site-directed mutagenesis was used to replace the disrupted gene. Residues that were predicted to be essential for C-6 hydroxylase specificity of OtcC were altered. Structure determination of the product made by this new strain revealed that it confers OTC production. This raises questions concerning either the specificity of the mutation or of the specificity of OtcC itself.
