Sažetak
A thin-layer chromatographic differentiation of the sphingolipid bases ceramides and cerebrosides, according to the number of the c=c double bonds, is described. The method is based on the substantial increase in polarity of the unsaturated lipid due to introduction, by oxidation with osmium tetroxide of two hydeoxy groups per double bond. The long-chain bases were N-acetylated before oxidation, whereas complex sphingolipids were oxidized without pretreatment. The difference in the Rf values (delta Rf) of unoxidized and the more polar oxidized material allows a clean-cut distinction of the long-chain bases and their complex derivatives (sphingolipids) with respecxt to the degree of unsaturation.
