Sažetak
A sensitive and precise liquid chromatographic method has been used for the determination of total thiamin, riboflavin and niacin content in three four types used in macrobiotic nutrition. The arbitrariy selected samples were extracted with hydrochloric acid (thiamin and riboflavin) and calcium hydroxide (niacin) in autoclave, followed by an enzymatic hydrolysis with takadiastase (EC 3.2.1.1). Thiamin was oxidized to thiochrom, while riboflavin and niacin were chromatographed in the original form. After cleaning procedure on C18 cartridges (Superclean LC-C18) the samples were chromatographed on a reverse-phase C18 column by using different combinations of mobile phase (methanol, acetonitrile, acetate buffer, phosphate buffer, respectively) and two detectors (UV Diode Array and Flourichrom). All the data obtained by LC technique showed no significant differences in comparison with the values obtained by AOAC fluorometric method and agreed with the vitamins levels claimed by the manufactures, as well.
