Celiac disease (CD) is an enteropathy mediated by intolerance to gluten. Commonly this disorder is multifactorial ; HLA genes and other genes together with environmental factors are involved in disease development. The aim of our study was to find out prevalence of DQ2 and DQ8 allele in celiac patients and first degree relatives. We collected blood from 83 person (27 celiac patients and 56 their first degree relatives). A group of 380 healthy unrelated and ethnically matched subjects were used as controls. Genomic DNA was extracted using standard method. HLA-DQB1 was determined by PCR-SSP method. The whole PCR reactions were directly electrophoresed in 2% agarose gels stained with ethidium bromide (0.5 mg/ml gel). The gel were run at 10v/cm for 20 min in 0.5X TBE buffer, visualized under UV light and documented by photography. Seventeen of 27 celiac patients (63%) had DQ2 allele (RR=4.055, CI 2.794-5.885, p<0.0001), four had DQ8 allele (15%) (RR=2.448, CI 0.912-6.570, p=0.0756). The prevalence of DQ2 in first degree relatives was 23 of 56 (41%) comparing with controls 59 of 380 (15.5%), p<0.0001. DQ8 was found in 11 family members (20%). The prevalence of DQ 8 in controls was 23 of 380 (6.1%), p=0.0013 HLA DQ2 is the most frequent allele among our celiac patients and first degree relatives. The prevalence of HLA DQ2 and DQ8 was significantly higher among first degree relatives than the controls.
