Sažetak
We investigated ATP-dependent transport of unconjugated bilirubin (3H-UCB) in vacuoles and vacuolar vesicles from the yeast Saccharomyces cerevisiae. In the presence of ATP, vacuoles take up UCB with a Km of 16.8 ą 2.7 nM and a Vmax of 190 ą 11 pmol UCB /mg prot per 45 sec. ATP-dependent UCB uptake was reduced by 60 ą 5 % and 38 ą 7% in YCF1 and YLL015 w-deleted cell vacuoles, respectively, while the double deletant strain showed no UCB uptake. On the contrary, taurocholate transport was fully comparable in vacuoles of wild-type and deletant yeast. These data identify YCF1 and YLL015w, hereby named BRT1, as the genes responsible for ABC protein-dependent UCB transport in the yeast.
