Sažetak
Carbamates belong to a group of compounds used in agriculture, public health and in clinical use. Some of the carbamates due to their high toxicity are used as chemical warfare agents. The mechanism of their action is based on cholinesterase inhibition, causing carbamylation of the catalytic site of this enzyme. The therapy of carbamates intoxication is limited to the use of atropine, because the oxime therapy is ineffective. Even more, the toxicity of carbamates increase when oximes are applied.
In this paper the evaluation of the therapeutic value of new oximes 1,1'-tetramethylene-bis-(2-hydroxyimomethyl-3-phenylimidazolium) dibromide (BDB-122) and 1-[(3-(2-hydroxyiminomethyl-3-methylimidazolio)-2-oxapropyl)-3-(N,N-dimethyl caramoyloxy) quinuclidinium diiodide (BM-1) in the intoxication of male mice with carbamates (propoxur, carbaryl, RO2-0683) is presented. The therapeutic effects against carbamate toxicity were tested by giving the oxime intraperitoneally (i.p.) at 1/4 of the respective LD50 dose together with atropine (10 mg/kg b.w.) immediately after subcutaneous (s.c.) administration of carbamate. Each oxime applied together with atropine had good therapeutic effect. Oxime BDB-122 given with atropine protected mice against 16.0 LD50 of propoxur, 2.0 LD50 of carbaryl, 8.0 LD50 of RO2-0683. Oxime BM-1 with atropine yielded a 100 % survival of mice intoxicated with 16.0 LD50 of propoxur, 2.5 LD50 of carbaryl and 20.0 LD50 of RO2-0683. Good therapeutic effect of tested oximes in the intoxication with carbamates is especially interesting, because actually, the drug of choice is atropine only.
