IL-6 has been implicated in tumorigenesis, however its role is still far from being clearly defined. Regulation of IL-6 expression is highly complex and additional complexity is introduced by SNPs in the IL-6 gene. These SNPs might influence mRNA transcription and create inter-individual differences in IL-6 expression which might in turn result in increased susceptibility to certain tumours. The aim of this study was to analyze IL-6 mRNA expression in sporadic colon cancer. Influence of IL-6 -174 G/C polymorphism on IL-6 mRNA expression and sporadic colon cancer susceptibility was evaluated as well. The frequency of IL-6 -174 G/C was analyzed by PCR-RFLP analysis in 160 patients and 160 controls. IL-6 mRNA expression in colon tumours and corresponding normal mucous tissue was analyzed by real time RT-PCR. There was no association of IL-6 -174 G/C with susceptibility to sporadic colon cancer. No statistically significant difference in IL-6 gene expression in tumour tissue compared with the corresponding normal tissue was observed (p=0.116). No correlation was found between IL-6 mRNA expression and histological grade or Dukes’ stage of tumours (p=0.508 ; p=0.978). There was no association of IL-6 -174 G/C genotypes with IL-6 mRNA expression in colon tumours and corresponding normal mucous tissue (p=0.355 ; p=0.152). Based on the results of our study we can conclude that IL-6 mRNA expression is not significantly different in colon tumours compared with normal mucous tissue. IL-6 -174 G/C polymorphism does not influence IL-6 mRNA expression in colon tumours and is not associated with altered susceptibility to sporadic colon cancer.
