Sažetak
Many investigations suport the hypothesis that macrophages play a vital function in tumor rejection. In previous experiments in our laboratory, we demonstrated that activated macrophages by water-soluble derivative of propolis (WSDP) are associated with resistance to tumor growth in CBA mice. Macrophages are able to destroy tumor cells under a variety of circumstance. To obtain more direct evidence regarding the role of WSDP on macrophage activity, we undertook the studies described below. Peritoneal macrophages from normal CBA mice and/or Swiss albino mice and mice injected with WSDP were examined for direct and indirect cytotoxic effect to tumor cells in vitro and in vivo. Direct cytotoxicity was estimated by measurement of incorporation of 3H-tymidine into HeLa cells or by colony forming ability of Hela or V79 cells in vitro. In addition, we measured the production of interleukin 1 (IL-1) and nitric oxide (NO) in culture of macrophages collected from mice treated with WSDP. Finally, we attempted to determine whether extracellular cytotoxicity to tumor cells is dependant on NO or other factors produced by actvated macrophages. The results established that antitumor activity of WSDP includes pronounced immunomodulatory activity due to the augmentation of non-specific antitumor resistance in mice via macrophage activation and production of soluble factors by those cells which may interfere with either cells of immune system or directly with by tumor cells
