Sažetak
In the process of pre-mRNA splicing in mammals, exon recognition by spliceosome is assisted by members of serine-arginine (SR) family of proteins, which specifically bind certain short regions in the exons, called exon splicing enhancers (ESEs). Loss of an exonic regulator can lead to skipping of that exon and formation of truncated protein. Degeneracy and context-dependence of these regulators makes biological evaluation of ESEs predicted by either of two existing in silico tools very difficult. I created ESE Analyzer Web Server for evaluation of predicted ESEs based on the alignments of human paralogous sequences and observation of ESEs conservation patterns, with special regard to competing selection pressures to protein versus enhancer sequences. Further, I performed extensive statistical analysis to discover the possible patterns regarding enhancers’ distribution in the human exons. I found no preferential density, position or distance between individual or clustered enhancers. I compared these parameters to the properties of the corresponding exons (length, splice site strengths) and detected no significant correlation.
