Sažetak
Phagocytosis by alveolar macrophages (AM) has been recognized beyond doubt as an important cellular first-line defense mechanism against inhaled particles that tend to deposit in the alveolar zone. This study was aimed at to quantitatively estimating of retention kinetics of various particles inhaled in the lungs and establishing relationship of particles retained in the alveolar compartment and at population of alveolar macrophages. This ratio was observed over a 150 days-lasting that followed the acute intratracheal instillation of 2.37mm in diameter wide polystyrene microspheres (2.4x108 microspheres/ml PBS) and asbestos fibres (0,625 mg asbestos and 1,2x108 microspheres/ml PBS) into the lungs of Wistar rats. Population of lung,s free cells obtained by bronchoalveolar lavage was analyzed both by light and fluorescent microscope determine the number of particles within those cells (i.e. the number of AMs containing 0, 1-2, 3-4....<16 particles/AM) in order to 1st, 7th, 30th, 80th and 150th postinstillation day. The percentages of AMs belonging to the lower particle burden categorie, (i.e. the AMs containing 1 or 2 particles per AM) increased from the first up to the 80th postinstillation day suggesting the fact that AMs with a low particles burden are not being removed from the alveolar zone. The efficiency of AM-clearance was altered during the whole study course, starting from the first postinstillation day. The results of the study revealed the distribution of inert particles within AMs during alveolar clearance to be gradually, while the retention of particles in the lungs of experimental animals instilled with toxic fibres shown to be merely retained in the interstitium.
