Sažetak
The aim of our study was to determine whether or not the F4ac fimbrial antigen of non-ETEC strain 1466 is capable of priming lymphocytes from jejunal lamina propria (JLP), ileal Peyer's patches (IPP), and mesenteric lymph node (MLN) of weaned pigs to respond to in vitro stimulation with Baypamun (BPM). Five 4-week-old pigs were orally inoculated with 10^10 CFU of either F4ac^+ (1466) or F4ac^- (1467) non-ETEC strains. The control pigs were given the medium only. At postinoculation Day 6 the cells were isolated and treated with BPM, and common mitogens (PHA or LPS) or peptidoglycan monomer (PGM), an immune response modifier (IRM), which were used as the control stimulators. The lymphocytes of 1467-inoculated pigs, with exception of those from IPP when stimulated with BPM, responded weaker to both mitogens than did the control cells. BPM also strongly stimulated (P < 0.01) the cells from IPP and MLN of pigs inoculated with strain 1466. Consistent with this is a better responsiveness of these lymphocytes when PHA (P < 0.05) or PGM (P < 0.01) where applied. However, the lymphocytes from JLP of 1466-inoculated pigs exhibited significantly lower (P < 0.01) responses to PHA or LPS than those of noninoculated controls. The results indicate that BPM exhibited properties of an IRM and, thus, could act sinergistically with tested vaccinal antigen (i.e., F4ac antigen of replicating E. coli strain 1466) in an attempt to stimulate the intestinal immunity against postweaning colibacillosis caused by F4^+ ETEC strains.
