An abortion is the most frequent complication of pregnancy with the incidence of 12-15% in population, and 25-60% demonstrated chromosome abnormalities. A retrospective and prospective study from 1983 couples with one or more spontaneous abortions is presented. Pedigree analysis and consulting with both parents (age, profession, smoking, alcohol, drugs, pets, earlier spontaneous abortions, stillborns, and other illnesses of both parents and their siblings), serological findings for several viruses, and cytogenetic and gene analyses were and would be performed. The purpose of this research is to confirm the hypothesis that two hits are responsible for spontaneous abortion. The first hit could have been triggered by patient's former diseases of urinary and/or genital system. The number of spontaneous abortions recorded in their siblings of second generation creates predisposition that may end with termination of pregnancy. The second hit is usually triggered by chronic presence of EBV virus in male or/and female partner. It has been demonstrated that there is no significant relation between the age of male of female partner s and the number of spontaneous abortions. Both males and females had former urinary or/and genital infections and the number of spontaneous abortions among the siblings of second generation is twice as high in relation to average population. Reactivation of latent EBV infection provides possibility for second hit. The second his causes a series of molecular and immunological changes that result with spontaneous abortions, as multifactorial trait.
