Sažetak
Abstract: In this work the processes of isolation and purification of laccase from Trametes versicolor were investigated. Fungus was cultivated in the form of mycelial pellets on the complex nutrient media consisted of the waste of pulp and paper industry. After cultivation, pellets were separated from enzyme suspension by the processes of filtration and centrifugation. Concentration of enzyme suspension was performed with ultrafiltration precess using ultrafiltration module (Ultrafiltration cell 8050, Amicon), where the enzyme suspension was concentrated 5 fold. The concentrate was purified using ion exchanger Sephadex G-25. The influence of temperature on efficiency of isolation process and on enzyme activity in the processes of ultrafiltration and ion exchange was investigated.
