Sažetak
Interactions between bovine serum albumin (BSA) and weak polyelectrolytes poly(allylamine hydrochloride) and poly(dimethylaminoethyl methacrylate) were investigated in solution and at interfaces as a model system for interactions between proteins and synthetic weak polyelectrolytes. Interactions at interfaces were examined by means of stagnation point optical reflectometry and electrophoretic mobility measurements and interactions in solution were investigated by potentiometric titrations. Application of stagnation point optical reflectometry for characterization of multilayers obtained by exposing a suitable substrate to solutions of a cationic homopolymer and an oppositely charged protein was tested. It was shown that protein concentration, pH and ionic strength influence the formation of polyelectrolyte-protein multilayers. Additionally, the adsorption of bovine serum albumin on previously formed polyelectrolyte multilayer was examined. It was observed that the adsorbed amount of BSA depends on the BSA concentration and that the introduction of the precursor layer also influences the adsorption process. The effect of salt on the behaviour of bovine serum albumin in solution and on the interactions between BSA and the weakly charged polyelectrolytes poly(allylamine hydrochloride) and poly(dimethylaminoethyl methacrylate) was investigated by potentiometric titrations. Titrations of pure BSA solution and of the BSA solution with added polyelectrolyte were performed in the presence of different salts. Three electrolytes having the same anion and a different cation were used, lithium chloride, sodium chloride, and caesium chloride. The titration curves showed a strong influence of ionic strength and the electrolyte type. The cation effect was found to be in accordance with the Hofmeister series.
