Objectives: The aim of this work was to determine the effects of diets having different n-6/n-3 ratio on lipid content and composition of organs highly involved in lipid metabolism. For that purpose, the reflection of diets enriched with pumpkin-seed oils (as a source of n-6 and n-9 fatty acids ), olive oil (n-9 fatty acids), lard (n-9 and saturated fatty acids) and fish oil (n-3 fatty acids), on lipid metabolism of mice liver, brain and epididymal adipose tissue was determined. The distribution of dietary fatty acids in tested organs and the changes of n-6/n-3 ratios were analyzed. Methods: Wild type C57Bl/6 male mice aged 8-10 weeks were fed for 3 weeks a standard diet (the control group) and diets enriched with 5% (w/w) of specific oil or lard. Total lipids from three tested organs were extracted according to Folch et al., while fatty acid composition of liver, brain and adipose tissue lipid fractions, as well as fatty acid profile of diets used in the study, were determined by gas chromatography analysis of the corresponding methyl esters. Results: Enrichment of the diet with specific oil or lard significantly changed the n-6/n-3 ratio, ranging from 1.18 to 18.88. In analyzed organs, ratio ranged from 0.77 to 3.98 in liver, 0.53 to 1.00 in brain and 8.35 to 16.10 in adipose tissue. Fish oil enriched diet provoked the highest reduction of the ratio (more than 4 fold) in the liver. Also, this type of diet significantly reduced n-6/n-3 ratio in adipose tissue. Surprisingly, the lowest n-6/n-3 ratio was noticed in the brain, in lard enriched diet. Total liver lipids of mice fed experimental diets ranged from 4.40 to 5.32%, while brain lipids were in a range from 7.3 to 8.52%, with the lowest values in both organs in diet enriched with fish oil. In adipose tissue total lipid content had the lowest value in fish oil enriched diet, and, interestingly in lard enriched diet. Conclusion: Presented data suggest that dietary fat composition induces changes in lipid fatty acids profiles, with liver tissue lipids as the most affected one. Fish oil enriched diet, which had the lowest n-6/n-3 ratio, provoked an accentuated reduction of the ratio in all tested organs, implicating the beneficial role of fish oil enriched diet in the prevention of many chronic diseases.
