Sažetak
Enzymes are widely used as biological catalysts because of their high specificity of action. It is well known that pancreatic lipase acting on triglycerides shows a positional specificity of hydrolysis since only the ester bonds in the external position od glycerol are split. Pancreatic lipase is solubilized in McIlvainen buffer solution (pH 6.0) in the presence of nonionics surfactant and water catalyzing the hydrolysis of triglycerides. Kinetic analysis of the lipase-catalyzed reaction was found to be possible in this system. The purpose of this work is to monitor the course of enzymatic hydrolysis in the presence of various concentrations of enzymes and various concentrations of substrates.
