Sažetak
The HLA-Cw typing using alloantisera is associated with a high frequency of blanks. In order to clarify these problem 106 unrelated healthy individuals were simultaneously tested using serology and one of DNA typing methods (PCR-SSP or ARMS-PCR). In 11 cases no Cw antigens were found by serology, while these samples were successful typed by DNA methods. Comparison between these two typing strategies in the rest of 95 individuals revealed a discrepancy of 24, 7% with the majority of these discrepancies due to serological blanks. Serology revealed blanks in 31.1%, whereas DNA typing revealed blanks in only 5, 8% cases. Most undefined serological specificities were Cw*12, Cw*17 (14, 2%). DNA typing was successful in all 7 (3, 7%) individuals in which serology failed to detect Cw*08. Moreover, DNA typing revealed incorrect serology typing in 3 individuals (3, 2%). A total of 144 unrelated Croatians (106 individuals tested simultaneously by serology and DNA typing and 38 individuals tested only by DNA typing) were analyzed for the frequency of Cw alleles in healthy population using ARMS-PCR (50 individuals) and PCR-SSP (94 individuals). Both methods have higher resolution than serology, but compared one to each other they have equal resolution. The most frequent alleles were as follows: Cw*0401 (20, 5%), Cw*0701 (10, 2%), and Cw*0602 (9, 7%). Comparison between our and other European populations did not reveal any significant difference. In conclusion, molecular typing is a method of choice in Cw typing which will enable to examine the role of Cw alleles in unrelated bone marrow transplantation as well as in disease association studies in the Croatian population.
