The CD26 molecule is a transmembrane glycoprotein with intrinsic dipeptidyl peptidase IV activity, present in a wide diversity of tissues and cell types, having multiple biological functions. It is expressed on activated lymphocytes T, B, NK-cells and macrophages. The cleavage of the membrane-bound DPP IV results in a soluble form that circulates in the serum and other biological fluids. It is involved in the activation and inactivation of numerous peptides of biological importance. Substrates of DPP IV are proline-containing peptides, which include growth factors, chemokines, neuropeptides, and vasoactive peptides. Therefore, it could play a variety of important roles in physiological and pathological processes. It has been suggested that DPP IV/CD26 is involved in the regulation of T lymphocyte migration to the site of inflammation in immune-mediated diseases. Changes in DPP IV/CD26 activity and expression have been observed in different chronic inflammatory diseases, likewise in autoimmune and malignant diseases. Here we review the current knowledge about the function and relevance of DPP IV/CD26 in the pathogenesis of chronic inflammatory diseases.
