Sažetak
Our objective was to investigate the role of good glycemic control in the progression of advanced glycation process. An immunoassay was developed to detect and quantify the AGEs formed in vivo in rat tendon and aortic collagen. Individual collagen samples were extracted by extensive pepsin and collagenase digestion. Aortic AGE-collagen content of untreated diabetic rats ((D)n=13, 184.9?12.3 AGE U/mg)was compared to diabetic rats treated for 12 wk with insulin ((DI)n=6, 133?10.7) ; phlorizin, an inhibitor of renal tubular glucose reabsorption ((DP) n=6, 132.4?8.9) or a combination of insulin and phlorizin ((DIP) n=6, 124.3?6.5. In spite of applied therapy, all groups of diabetic animals had a significantly higher aortic AGE-collagen content than the control group rats (C, n=8, 104.6?14.9)of the same age (D, DI, DP, DIP vs C, p?0.001). Comparison between the mean levels of HbA1c (D:5.62?0.38 vs C:1.7?0.05%) and mean AGE levels yielded a very good exponential correlation (r=0.89, p?0.001). SDS- PAGE showed higher molecular weight fragments in diabetic collagen samples, pointing to cross- linking. Our study provided immunochemical evidence for AGE in vivo formation and their relation to diabetes duration. Glycemic control by insulin or independent of it retards and reduces the intensity of advanced glycation process without abolishing it. Therefore, additional therapeutic forms appear to be required.
