Sažetak
The aim of the project is to develop a method for determination of percutaneous penetration of polyethylene glycols (PEGs) of different molecular weight (MW) through the stratum corneum (SC). The method will be used to study the differences in percutaneous penetration between normal and impaired skin (e.g. by patients with atopic dermatitis or by skin impaired with sodium lauryl sulfate) in respect to MW. PEGs are extensively used in research of intestinal permeability1. The water/octanol partitioning (Po/w) coefficient doesn’ t change greatly with molecular mass which justifies their use as appropriate molecular size model compound not compounded by lipophilicity. Skin penetration of PEGs has already been studied in vitro in murine skin where the dependence of permeability on MW with different forms of barrier disruption has been demonstrated2. This is for the first time that percutaneous penetration of PEGs has been determined in human skin in vivo. The water mixture of PEG containing different oligomers of MW of 150-700 Da was applied onto the lower forearm using round Finn Patch test chambers with a diameter of 18 mm for 6 and 24 hours. 15 minutes after the end of exposure, pieces of adhesive tape (precut 18 x 18 mm2, Diamond Ultra Clear Tape) were consecutively applied to the test sites. The tapes were fixed to the skin by pressing with a stainless steel roller of 1 kg. Tapes were consecutively applied until the SC is totally removed as observed by the shinny appearance of the skin and by measuring the trans-epidermal water loss (TEWL). The analysis of PEG oligomers in subsequent tape strips is based on derivatization with petafluoropropionic anhydride (PFPA) and gas chromatography-electron capture detection (GC-ECD). The amount of the proteins in each tape strip was analyzed spectrophotometricaly by modified method of Dreher3 et al. The protein analysis was used to measure the amount of the SC removed by a successive strip, and to assess the depth of the consecutive SC strip. In this study we quantified both, model compound and proteins on the same tape strip, thus allowing normalization of the PEG concentration for protein amount. The results were expressed in amount of each oligomer of PEG /amount of proteins versus SC depth. The developed method allowed us to measure the concentration of PEGs with MW up to 650 Da. The preliminary results showed that the MW cut off is shifted to higher MW in the skin pre-treated with SLS and that the amount of PEGs penetrated the SC was significantly higher than in non-treated skin.
