Sažetak
The acute effect of selective 5-HT1A agonist flesinoxan on the 5-HT synthesis rate was investigated in discrete rat brain regions, using an autoradiographic method (Diskic et al., J Cereb. Blood Flow Metab., 15:806, 1995), in order to get a better understanding of the role of 5-HT1A receptors in the brain serotoninergic system. Flesinoxan (5 mg/kg) decreased concentrations of plasma free tryptophan. In 24 out of 27 brain regions studied flesinoxan induced significant decrease in the 5-HT synthesis rate. In the areas of serotoninergic cell bodies the decrease in flesinoxan-treated rats, calculated as a percent (%) of the synthesis rate in the control group (saline treated rats) was -50%, -48%, and -32% in the raphe dorsal, median and magnus, respectively. After flesinoxan administration the decrease in the 5-HT synthesis rate in the areas with high density of postsynaptic 5-HT1A receptors (hippocampus: -28%; superior colliculus: -36%) was similar to decrease in synthesis rate in the regions with low density of postsynaptic 5-HT1A receptors (substantia nigra: -41%, hypothalamus: -39%; dorsal thalamus: -37%; lateral geniculate body: -37%). Our results suggest that flesinoxan induced changes in the 5-HT synthesis rate are mediated through activation of both somatodendritic and postsynaptic 5-HT1A receptors.
