Sažetak
Human papillomaviruses (HPV) are a heterogenous group of viruses, which consist of more than 70 genotypes. They cause benign tumors, precancers and cancers of the skin and mucosa. They are usually found in low- and high-grade cervical intraepithelial neopasia and other urogenital regions (vagina, anus, penis, prostate). In this study we determined the presence of HPV in colon adenocarcinoma. Thirty six patients with sporadic colon carcinoma were included in this study. For each patient a tumor tissue and adjacent normal mucosa were collected. After DNA extraction, consensus polymerase chain reaction (PCR) with degenerative HPV L1 MY09/MY11 (Cancer Cells 7, 209-214, 1989) and LC (Int J Cancer 45, 990-992, 1990) primers was performed. Since no positive result was obtained, the sensitivity of the method was increased by using nested PCR with GP5/GP6 primers (J Gen Virol 71, 173-181, 1990) within MY09/MY11 PCR product. By using nested PCR, HPV DNA was detected in 5 patients ; in one case in both tumor and normal tissue (Dukes' C), in three cases only in tumor (Dukes' A, B and C) and in one case only in normal tissue. In those samples a type-specific PCR for HPV genotypes 6, 11, 16, 18, 31 and 33 was performed and no positive results were obtained. The presence of HPV DNA was determined in 5/36 (13.88%) colon adenocarcinoma. Since the positive results was obtained only with highly sensitive nested PCR, it can be assumed that HPV is present in low-copy number in colon cancer cells. The fact that HPV genotype could not be determined can be explained either by presence of HPV types other than 6, 11, 16, 18, 31 and 33 or by lower sensitivity of type-specific PCR compared to nested PCR approach.
