Sažetak
V. ammodytes ammodytes venom has been used for active immunisation of animals in antivenom production. We have found recently that the protective efficacy of antisera from venom-immunized rabbits highly correlated with the level of IgGs specific for Ammodytoxin A (AtxA)1. It has also been shown that higher Atx content contributes to higher lethal toxicity of the venom sample2. Also, the venom with higher Atx content induced higher level of anti-AtxA antibodies2. Obviously, the in vitro methods determining Atx content might be predictive of the toxicity and immunogenic potential of venom samples. The aim of this work was to test the two in vitro methods for AtxA content determination: a) recently described chromatographic method2, and b) the novel sandwich ELISA, and to compare results obtained for panel of several different venom samples with their lethal toxicities (LD50) determined by in vivo assay. We also compared the results of the novel assays to the results of several methods currently routinely used for snake venom characterisation (MHD, MND, SDS-PAGE, protein content determination). The results obtained showed that new methods might allow quick and reliable screening of the AtxA-content in the venom sample before using it for the immunisation of experimental animals for antiserum production. 1Halassy et al.
