For several years two systems have been in use for the numbering of the paraoxonase (PON) amino acid residues: 1. designating the initiation site methionine as number one as suggested by Hassett C., 2. starting with the first residue (alanine) of the mature human serum PON1 as proposed by Adkins S.. The expanding number of PON-like genes discovered, and cDNA sequences of the PON family being added to the gene data banks in recent years are important reasons to try to resolve this problem. An additional reason are clinical studies dealing with the human PON1 and PON2 polymorphisms, and the incidence of various diseases, or with organophosphate toxicity. Because of the post-translation modifications of PON the first amino acid from N-terminus was removed and the numbering of amino acid residues is changed. The only common denominator among all PON family will likely be their initiation methionine residues. An obvious advantage of using the initiation methionine residue number one is that it can be the reference point for all members of PON family, regardless of how they are processed, or what amino acid turns out to be the N-terminal residue.
