Sažetak
The objective of the present work was to monitor the involvement of oxidative stress caused by tumour growth on renal calcification and development of 4-hydroxy-2-nonenal (HNE) protein adducts. Therefore, we used rat Walker 256 carcinoma (W256) an animal model for humoral hypercalcaemia of malignancy (HHM) on Wistar rats. Some animals were additionally treated with Sephadex which causes an aseptic inflammation absorbing in a subcutaneously formed papula activated neutrophils and inflammatory exudate. Upon death of animals kidneys were surgically removed and analysed for HNE-His adducts and renal calcifications. The distribution of HNE-His adducts was analysed by immunoperoxidase technique. The changes in renal cortex calcifications followed the onset of HNE-immunopositivity, thus suggesting that the development of HNE-protein adducts may be associated with development of renal calcificates in tumour bearing organism. Treatment with Sephadex, which disctracts inflammatory granulocytes responding to W256 from tumour into aseptic subcutaneous papula reduced for more than 35% the presence of kidney calcificates. Therefore, the results of this study imply that the development of renal calcificates due to HHM may be associated with the systemic inflammatory response to cancer development and consequential oxidative stress generating HNE-protein adducts in kidney.
