Sažetak
Due to high frequency and fatality malignant diseases are one of the main issues of medicine. A lot of attention is given to the natural compounds because they can interfere with tumor growth. Caucalis platycarpos L. was used in folk medicine for the treatment of certain types of tumor. Water extract of the aboveground parts of this plant showed a remarkable antitumor activity in rats and mice. However, chemical composition is only partially defined from general phytochemical and chromatographic investigations. A method coupling high-performance liquid chromatography (HPLC) with diode-array detector (DAD) and electrospray ionization mass spectrometry with ion trap analyser was optimized for the separation and identification of phenolic acids, flavonoid glycosides and flavonoid aglycones in the extracts of C. platycarpos L. HPLC experiments were performed on ZORBAX SB-C18, 4.6x250 mm, particle size 5 μ m with suitable guard column. The mobile phase consisted of 10 mM ammonium formate in water-acetonitrile 95:5, pH = 4.0 (A) and 10 mM ammonium formate in water-acetonitrile 50:50, pH = 4.0 (B). The gradient elution started with 0% B and changed to 40% B in 65 minutes, then reached 100% B in 35 minutes. The flow rate was 1.0 mL/min and split out 200 μ L/min to MS. All mass spectrometry data were recorded in the negative ion mode. The screening was performed in full scan, auto MSn, MRM and manual MSn modes to fragment the major ions obtained in every step. Commercially available standards were used where possible. Chlorogenic acid, luteolin-7-glucoside, apigenin-7-glucoside, quercetin-3-galactoside, luteolin and apigenin were identified comparing experimental data for retention time (tR), UV and MS spectra with those of the authentic compounds. Chrysoeriol-7-rutinoside was isolated by preparative liquid chromatography and identified after elution by HPLC/MS, by direct injection into the MS system and 1H NMR. Luteolin-7-rutinoside, apigenin-7-rutinoside, isomers of dicaffeoyl quinic acid and caffeoylferuloyl quinic acid were identified by comparing experimental data for UV and MS spectra with those of literature. Above mentioned flavonoid glycosides and phenolic acid esters, as well as isolated chrysoeriol-7- rutinoside, are identified as constituents of C. platycarpos L. for the first time.
