Sažetak
Department of Pathology and Cytology, University Hospital Merkur, Zagreb, Croatia Aim: Tumor cells of DLBCL are derived from germinal centers or post germinal centre B-cells. Recurrent genetic abnormalities and the association between high protein expression levels and poor prognosis have indicated that FOXP1 has a role in DLBCL tumorigenesis. FOXP3 has a prognostic significance in folllicular lymphoma ; therefore it could have a role in lymphomas derived from the same cells of origin. We report experiences investigating FOXP1 expression and genetic abnormalities in DLBCL from a single hematopathology centre in Croatia, as well as evaluation of FOXP3 positive cells in DLBCL. Patients and methods: 53 samples diagnosed as de novo DLBCL were CD10, MUM1, BCL6, BCL2, FOXP1 and FOXP3 immunostained. FISH analysis was used for the detection of gene aberrations involving FOXP1, BCL2 and BCL6 loci. Data were compared using χ 2 test. Results: FOXP1 protein was detected in 28 cases, aberrations involving the FOXP1 locus were found in 19 cases with 13 cases showing both events simultaneously. FOXP3 positive cells were detected in 37 cases. χ 2 test showed a statistically significant correlation of BCL2 expression and FOXP1 genetic abnormalities as well as BCL2 expression and BCL2 genetic abnormalities. Correlation was also observed between BCL6 and FOXP1 genetic abnormalities. Conclusion: FOXP1 is expressed in Croatian DLBCL patients and shows varying levels of expression. FOXP3 positive cells are present in DLBCL. We present data that confirm a connection between FOXP1 and BCL2 in DLBCL in patients from a single centre, as well as lack of FOXP3 positive cells impact on tumor development.
