Sažetak
The mumps virus (genus Rubulavirus, family Paramyxoviridae) is an enveloped, non-segmented, negative strand RNA virus with the genome size of 15 384 nucleotides. The virus genome contains seven genes in the following order on the genome map: the nucleocapsid (N), phospho (P), matrix (M), fusion (F), small hydrophobic (SH), hemagglutinin-neuraminidase (HN) and large (L) protein gene. Besides phosphoprotein, P gene encodes two more proteins, V and I. Mumps is an acute communicable disease in humans, the only natural host of mumps virus. The virus infects numerous organs, including central nervous system. Data regarding the pathogenesis of MuV are rare and the molecular basis of MuV attenuation/virulence is not clear. It is known that some strains are more virulent than the others. The virulence is not only a property of wild type viruses. It also represents a problem for attenuated vaccine strains due to the development of neurological side effects. Virus rescue, reverse genetics, or de novo synthesis of non segmented negative-sense RNA viruses from cloned cDNA is a reliable technique to study this group of medically important viruses. The system is based on intracellular transcription of viral full-length RNAs and simultaneous expression of viral proteins required to form the viral ribonucleoprotein complex (RNP). We intend to construct a RNA polymerase 1-driven rescue system of various mumps virus strains in order to study the role of different viral proteins in various aspects of the virus life cycle as well as the roles of virus proteins in virus-host interplay and pathogenicity.
