Sažetak
Transduction is biochemical technique for the introduction of full-length proteins into the cells. This process occurred in a rapid, concentration dependent fashion that is independent of receptors and transporters. Transduction of peptides and proteins has a broad implication in experimental systems for regulating intracellular processes. It also has a potential to be used in development of new therapeutic strategies for cancer therapy. Hyper proliferation of cancer cells is associated with deregulation of cell cycle progression, which is driven by the activities of CDKs. A key regulator of their activities is protein p27. It has significant role in cancer progression and antitumor drug response. The correlation between low p27 expression and either high tumor grades or poor survival has been reported. Conversely, it’ s over expression in cancer cells inhibits tumor cell growth by enforcing cell cycle arrest and apoptosis. To examine p27 as specific target molecule and its role in tumor cells apoptosis, transduction of TAT-p27, TAT-ptp27 and TAT-N'p27 was performed. Some transduced proteins influenced the expression of cell cycle proteins and induced apoptosis in tumor cells. Protein transduction therefore could give an opportunity for delivering of drugs into cells with emphasis on specific target molecules.
