Sažetak
Our objective was to determine relationship of advanced glycation process with glycemic control and duration of diabetes. Competitive ELISA was developed to quantify AGEs formed in vivo in rat tendon and aortic collagen. Individual collagen samples were extracted by extensive pepsin and collagenase digestion. Results are expressed as AGE U/mg collagen. Diabetic rats showed a significant increase in AGE-aortic collagen at 20 wk (n=6, 206.6ą16.7) compared with that at 4 and 12 wk (n=6, 110ą12.8 ; n=13, 184.9ą12.3 ; p<0.001). To investigate the influence of glycemic control on AGE-content we compared aortic AGE-collagen in untreated diabetics ((D) n=13, 184.9ą12.3) with rats treated for 12 wk with insulin ((DI) n=6, 133.9ą10.7), phlorizin, an inhibitor of renal tubular glucose reabsorption ((DP) n=6, 132.4ą8.9) or by the combination of insulin/phlorizin ((DIP) n=6, 124.3ą6.5). All groups of diabetic animals had in spite of the applied therapy, a significantly higher aortic AGE-collagen than controls (C: n=8, 104.6ą14.9) of the same age (D, DI, DP, DIP vs C, p<0.001). SDS-PAGE showed higher molecular weight fragments in diabetic collagen samples. Our study provided immunochemical evidence that AGEs are created in vivo, and are related to duration of diabetes. Glycemic control by insulin or independent of it retards and reduces the intensity of advanced glycation process without abolishing it.
