Sažetak
A protocol based on multiple stage tandem mass spectrometry (MS) for systematic and thorough investigation of the ganglioside species in human brain was developed. The unique feature of the high capacity ion trap (HCT) multiple stage MS to provide efficient dissociation, ultra-high reproducibility, sensitivity, throughput and accurate structural data, offered a reliable structural characterization of gangliosides GM1 and GD1 present in ganglioside mixture isolated from fetal neocortex ; GM1a as well as GD1a positional isomers were clearly identified. For detailed structural analysis of gangliside species, multiple stage MS enabled stepwise fragmentation at high sensitivity in up to 4 sequencing events.
