Sažetak
Different human diseases are characterized by inflammation and serve fibrosis (i.e. atherosclerosis, liver fibrosis..) in which an overexpression of the proapoptotic TGF-β 1 is often accompanied by the increase of an antiproliferative aldehyde, 4-hydroxynonenal (HNE), a lipid peroxidation end-product. In our laboratory we observed a cross-talk between the pathways mainly activated by TGF-β 1 (Smad proteins) and HNE (JNK signals). During STSM in Zagreb Rudjer Boskovic Institute, similar experiments were performed on primary cultured cells derived from the human colon adenocarcinoma tissues: HNE is able not only to switch on both JNK and Smad, but also to amplify these two signals. Therefore, we wanted to verify the hypothesis of an interaction between these two molecules on the cell growth control: in particular, we tried to see if there was a correlation between tumor deviation, HNE content and TGF-β 1 signal activity. These evaluations were performed by immunohistochemistry in human biopsies at different stages of dedifferentiation, i.e. normal tissues, tubular adenomas, villotubular low and high grade adenomas, Dukes A, B and C carcinomas. The immunohistochemistry revealed a gradual decrease of the aldehyde tissue content and a derangement of TGF-β 1 signal: the concentration of TGF-β 1 receptor II, which binds the cytokine and trigger the TGF-β 1 signaling cascade, was lowered proportionally to the grade of malignancy and the HNE staining.
