Sažetak
The cholinesterase inhibitor neostigmine produces analgesia epidural administered in human patients1 and intrathecal administered in sheep2. The aim of this study was to compare analgesic effects of two epidural administered drug combinations in sheep. Twelve adult cross-breed ewes were used in this study. Sheep were randomly assigned to one of two treatment groups. The lidocaine (4 mg kg-1) and neostigmine (15 µ g kg-1) was administered in group 1 and lidocaine (4 mg kg-1) and morphine (0, 1 mg kg-1) in group 2. The administration was performed epidurally in the lumbosacral intervertebral space before surgery. During general inhalation anaesthesia, arthrotomy was done on the right stiffle and articular cartilage defect was created. Blood samples for serum cortisol determination were taken before surgery and 12, 24 and 48 hours after surgery. Analgesia was evaluated subjectively with a numeric ranking scale that included assessments of comfort, movement, flock and feeding behaviour, respiratory rate, soft tissue swelling and response to wound palpation 8, 16, 24 and 48 hours after surgery. Ketoprofen was administered when a score > 9 (maximum score 24) was achieved. Data were analyzed using ANOVA and Duncan's test (p<0, 05 significant). Serum cortisol concentrations were significantly higher in the group 1 as compared with the group 2 at 12, 24 and 48 hours after surgery (p<0.05). Comfort and respiratory rate were significantly higher in the group 1 at 8 hours after surgery. In this study epidural lidocaine-morphine induced a higher and lasting analgesic effects than lidocaine-neostigmine.
