Sažetak
The aim of this study was to determine the in vitro effect of RoekoSeal (RSA), silicone-based endodontic sealer, on cytokine IL-1 and IL-6 production in mice macrophages. Freshly mixed sample of RoekoSeal was placed in contact with cultured mice macrophages. The supernatant from the cultures was assayed for IL-1 and IL-6 cytokine using enzyme-linked immunosorbent assay. Cells cultured in the absence of tested materials served as negative controls. As a positive control, similar cultures were also exposed to lipopolysaccharide. The silicone based material examined in these experiments did not stimulate increased IL-1 and IL-6 release from cell lines. The difference between IL -1 production by macrophages exposed to the tested materials and IL-6 production by the same macrophages was tested by one-way ANOVA. To compare LPS stimulated IL -1 and IL -6 production in the positive control group and to compare IL-1 and IL-6 production in negative control group were used Man-Whitney test. The silicone based material did not stimulate increased IL-1 and IL -6 release from cell lines examined.
