Background and purpose: New potential adjuvants, adamantyltripeptides (AdTPs, from the groups of muramyl dipeptide derivatives), were previously shown to be potential immunomodulators and antimetastatic agents. The purpose of this study was to investigate the in vivo effect of AdTPs on mice spleen lymphocyte populations, measured by flow cytometry. Materials and methods: Flow cytometry analysis of mice spleen lymphocytes was performend 24 hrs following single i.v. or i.p. administration of different doses of AdTPs (AdTP1 and AdTP2) and peptidoglycan monomer (PGM) used as a referral compound. Results were expressed as a percentage of cells positive for CD3 (total T-lymphocytes), CD25 (IL-2R), IaK (B-lymphocytes) and CD23 (activated B-lymphocytes) surface antigens. Results and Conclusions: We have shown that distribution of cells among lymphocyte population in the mouse spleen has been already affected after single administration of AdTPs. Two stereoisomers of adamantyltripeptide showed a different in vivo effect on mice spleen lymphocytes. Percentage of total T-lymphocytes (CD3+) was decreased, particularly subpopulation of cytotoxic T-lymphocytes (CD8+), after i.v. administration of AdTP2, but not with AdTP1. After i.p. administration of tested compounds, AdTP1 increased percentage of T lymphocytes (CD4+ and CD8+), while AdTP2 increased percentage of total (IaK+) and activated (CD23+) B-lymphocytes. PGM showed the same effect as AdTP2 in all experiments. Different effects of two tested stereoisomers (AdTP1 and AdTP2) on mice spleen lymphocyte should be further evaluated.
