Sažetak
Lysis of cells is accomplished using a motorised glass/teflon homogeniser. During the homogenizing procedure the samples should be kept constantly cooled (on ice) and all buffers added should be cold. Beta-mercaptoethanol is added to protect protein thiols from oxidation. The supernatant obtained after centrifugation at 50000 x g for 120 min is called the S50-fraction and contains the cytosol.
