Sažetak
Reactive oxygen species (ROS) are generated during oxidative metabolism. They can damage all macromolecules and finally cause cell death. Generaly, term oxidative stress is used to describe the level of oxidative damage in a cell, tissue or organ, caused by ROS. Three factors can induce oxidative stress in cells: increase in oxidant generation, decrease in antioxidant protection or failure of repairing oxidative damage. Oxidative stress is not a disease, but a condition that can lead to, or accelerate it. Enzymes of peroxidase family are oxidoreductases, what means that they catalyze oxidation/reduction reactions. Superoxide dismutase, catalase and gluthation peroxidase are examples of antioxidant enzymes that are produced by organisms. By measuring total peroxidase activity and protein concentration in the hemolyzed human red blood cells, I investigated the correlation between enzyme activity and age, for each sex separately, as well as for both sexes together. Total peroxidase activity has been analyzed spectrophotometrically at wavelength of 470 nm using substrates hydrogen peroxide and guaiacol. Peroxidase activity produces tetraguaiacol, a brown colored product. The intensity of color developed is proportional to enzyme activity. Total protein concentration has been determined by method of Lowry and used for enzyme specific activity calculation. The results obtained display higher total peroxidase activity in hemolyzed red blood cells of men compared to women. Difference between men and women in specific peroxidase activity and protein concentration is not significant. For both sexes statistically significant correlation has been proved between peroxidase activity and age. Our conclusion from the presented results is that ageing is accompanied by lowering of enzyme antioxidant activity in human red blood cells hemolyzate.
