Coumarins and hydroxycoumarin derivates have attracted intense interest according to their diverse pharmacological activities. Their role in plant as in animal cells is diverse. The purpose of this work is to investigate the effect of 3-(2-bromine-1-oxso)-7-bromine-4-hydroksycoumarin dissolved in proper solvent on the total peroxidase activity of hemolyzed erythrocytes. Several individual hemolyzed men blood samples of certain age group (know to have high peroxidase activity) were gathered to form a basic sample used in peroxidase activity measuring. Total peroxidase activity was recorded spectrophotometrically. As substrates, besides hydrogen peroxide, we used guaiacol which changes color in brown in the presence of peroxidase. The amount of color product, tetraguaiacol, was quantified by measurement at 470 nm. Increase in 3-(2-bromine-1-oxso)-7-bromine-4-hydroksycoumarin concentration in the proper solvent, induces gradual decrease of total peroxidase activity. Even the small concentrations of 3-(2-bromine-1-oxso)-7-bromine-4-hydroksycoumarin in the proper solvent cause fast decrease of peroxidase activity. Maximum inhibition is achived by 0.125 mM concentration of 3-(2-bromine-1-oxso)-7-bromine-4-hydroksycoumarin, independently on the used solvent. 0.125 mM K1 in ethanol is inhibiting 57.1% of peroxidase activity, while 0.125 mM K1 in DMSO is inhibiting 100% of peroxidase activity. IC50 value for3-(2-bromine-1-oxso)-7-bromine-4-hydroksycoumarin is 0.08 mM and 0.058 mM for ethanol and DMSO, respectively. 96% ethanol and DMSO were used as solvents for 3-(2-bromine-1-oxso)-7-bromine-4-hydroksycoumarin. The effect of each solvent on the peroxidase activity was analyzed separately by using only substrate, blood sample and applied solvent in the incubation mixture. Small amounts of used solvents significantly decrease total peroxidase activity. Maximum inhibition is achived using 20 μ L of ethanol, with peroxidase activity inhibited by 73%. Using 200 μ L of DMSO 64.3 % of peroxidase activity is inhibited. I concluded that the applied model of enzyme assay and used solvents are suitable for analyzing inhibitory effects of 3-(2-bromine-1-oxso)-7-bromine-4-hydroksycoumarin.
