Sažetak
Galectin-3 and CD44v6 have been proposed as tumor markers that could improve accuracy of preoperative diagnosis of thyroid lesions. In this study we wanted to find if the presence of macrophages and Hurthle cells in different types of benign thyroid lesions could provide an explanation for false positives by RT-PCR method. RT-PCR analysis of galectin-3 and CD44v6 expression was performed on RNA isolated from aspirates obtained by ultrasound guided fine-needle aspiration biopsy from 123 unselected patients with benign thyroid lesions. Results of RT-PCR analysis were evaluated against the definitive fine-needle aspiration cytology diagnosis or against the histological findings. The presence of macrophages and Hurthle cells in aspirates was evaluated as: absent (0), few (1) or abundant (2). Galectin-3 expression was found in: 5 of 17 (29%) follicular adenoma samples (all histologically verified), 7 of 27 (26%) Hashimoto thyroiditis samples, and in 19 of 79 (24%) nodular goiter samples. Macrophages were absent in 77% of galectin-3 negative cases as compared to 10% of galectin-3 positive cases in nodular goiter samples (p<0.01). Similar situation was found in Hashimoto thyroiditis samples: macrophages were absent in 75% of galectin-3 negative cases as compared to 14% of galectin-3 positive cases (p<0.01). We did not find a statistically significant (p>0.01) relation between the presence of Hurthle cells and galectin-3 positivity in benign thyroid lesions. CD44v6 expression was found in 5 of 17 (29%) follicular adenoma samples, 9 of 27 (33%) Hashimoto thyroiditis samples, and in 14 of 72 (18%) nodular goiter samples. Hurthle cells were absent in 94% of CD44v6 negative cases as compared to 50% of CD44v6 positive cases in nodular goiter samples (p<0.01). In Hashimoto thyroiditis samples Hurthle cells were absent in 50% of CD44v6 negative cases as compared to 11% of CD44v6 positive cases (p<0.05). We did not find a statistically significant (p>0.01) relation between the presence of macrophages and CD44v6 positivity in benign thyroid lesions. In follicular adenoma samples, we did not find a statistically significant (p>0.05) relation between the presence of macrophages or Hurthle cells and galectin-3 or CD44v6 positivity. We found a statistically significant relation between the presence of macrophages and galectin-3 positivity as well as the presence of Hurthle cells and CD44v6 positivity in nodular goiter and Hashimoto thyroiditis samples.
