Sažetak
Problem: Non-classical MHC-I molecule HLA-G, selectively expressed on invading extravillous cytotrophoblast, is in close contact with decidual CD56+ NK cells infiltrating first trimester decidua. We showed recently that exposure to HLA-G positive trophoblast altered their phenotype and function (decidual NK cells were able to induce FasL protein expression in close contact with HLA-G molecule and it represented dominant mechanism of cytotoxicity for K562-HLA-G cells). Therefore, our aim was to investigate further influence of this molecule on decidual NK cells. Methods of study: Decidual tissue was minced, treated with collagenase and passed through density gradient. After centrifugation, decidual mononuclear cells were collected. Intracellular expression of cytolytic mediators, NK cell activating receptors and Th1 cytokines was determined in decidual CD56+ cells after their 18 hour-culture with K-562-PWD cells and K-562-HLA-G transfectants at 37 C (1:1). Immunohistology for FasL and TRAIL was performed. Results: After overnight culture, decidual NK cells decreased their content of perforin in close contact with HLA-G molecule, comparing to their co-culture with K-562-PWD. We also observed HLA-G mediated decrease of activating NK receptor expression (NKG2D, NKG2C, NKp44, NKp46) as well as Th1 cytokine expression (IFN- , IL-18, TNF- ) in decidual NK cells. FasL and TRAIL were detected on paraffin-embedded first trimester decidual tissue sections as well as FasL by using Alexa 555. Conclusions: HLA-G decreased expression of cytolytic mediators, NK cell activating receptors and Th1 cytokines in decidual CD56+ cells. Since expression of HLA-G molecule is limited to gestation, it seems to be important in the regulation of various decidual NK cell functions.
