Sažetak
Before gastrulation mammalian embryos respond to external deleterious factors according to "all or none rule". After the onset of gastrulation this plasticity of development is lost and malformations are present at birth. To explore possible teratogenic action of different substances in vivo or in vitro systems are available.Until recently only preimplantation embryos were cultivated in serum-free media. We have succesfully cultivated, in several simple serum-free media, Fisher rat embryos isolated at the period of gastrulation.After grafting previously cultured embryos to a richer environment under the kidney capsule, a higher degree of differentiation was achieved. Nevertheless, 14 days in proteinless culture medium previous to grafting still restricted differentiation, especially of neural tissue. Therefore a conclusion can be drawn that our in vitro serum-free model might be sufficient to asses the impact of external factors upon mammalian embryo.
