Sažetak
This introductory lecture will discuss the basic principles of mass spectrometry (MS). The brief overview of ionization methods, such as electron ionization (EI), chemical ionization (CI), fast atom bombardment (FAB) and laser desorption (LD) ionization will be given. Two ionization techniques convenient for ionization of non-volatile compounds with molecular weights exceeding hundreds of thousands of daltons e.g. electrospray (ES) and Matrix- assisted Laser Desorption Ionization (MALDI) will be discussed in more details. The emphasis will be placed on the sample preparation and the selection of suitable matrices for the analysis of different analytes. Once the ions have been produced, they have to be separated according to their masses. Scanning analyzers (quadrupole or magnetic sector instruments) transmit ions of different masses successively along a time scale, while the others (time-of- flight mass analyzer, ion trap and ion cyclotron resonance instruments) allow the simultaneous transmission of all ions. The mass analyzers will be compared according to their main characteristics: the upper mass limit and resolution. Since the time-of-flight spectrometer (TOF) is well suited to the pulsed nature of laser desorption ionization, MALDI/TOF is widely applied in analysis of macromolecules. Linear time-of-flight mass spectrometer as well as tandem mass spectrometry with TOF mass analyzer will be described.
