Sažetak
Sperm are particularly susceptible to oxidative stress. Spermatozoa are protected by various antioxidative molecules in seminal plasma and in the spermatozoon itself that prevent excessive formation of reactive oxygen species. The objective of the present study was to examine age-related changes of antioxidant enzymes, glutathione and oxidation of lipids and proteins in bull spermatozoa. Semen was collected by artificial vagina from two groups of Simmental bulls: eight young bulls aged between 2.5 and 4 years, and seven older bulls aged between 5 and 10 years. In the spermatozoa of both groups of bulls spectrophotometricaly were determined the activities of glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD), as well as concentrations of reduced glutathione (GSH) and lipid peroxides (TBARS) and protein carbonyl content (PCC). Young bulls had significantly higher activity of SOD than the group of older bulls (2311.65± ; 354.59 U/g protein versus 1403.82± ; 278.09 U/g protein, respectively ; mean± ; SEM ; P<0.03). In the spermatozoa of younger bulls the PCC (0.32± ; 0.02 mmol/g protein) were significantly higher (P<0.01) than in the spermatozoa of older bulls (0.26± ; 0.02 mmol/g protein). Although the mean GSH concentration in spermatozoa of the younger bulls was higher (0.83± ; 0.08 micromol/g protein) in comparison with GSH in the older bulls (0.61± ; 0.10 micromol/g protein), no statistically significant differences were found between the groups (P>0.07). The GSH-Px activities and lipid peroxide concentrations in spermatozoa also did not significantly differ between the groups. This study demonstrated that the spermatozoa of the younger bulls are more susceptible to protein oxidation than those cells of older bulls and have higher SOD activity.
