Sažetak
The psychopharmacs, Moditen and Haldol has been widely used in the treatment of psychiatric disorders. The are conflicting results about genotoxicity of these drugs. To evaluate a possible genotoxic activity of psychopharmacs we performed comparative investigations using Ames salmonella/microsome test (strains TA100 and TA98), Allium test and human peripheral blood lymphocytes in culture in vitro. Plate incorporation assay with S. typhimurium demonstrated direct mutagenicity of Moditen (base substitution) whereas Haldol was ineffective. In Allium test both drugs caused a changes in the structure of chromosome material. It was found out that tested psychophormacs induced the occurrence of binuclear cells and changes on interphase nuclears.
Analysis of human peripheral blood cells showed that these drugs caused forming of binuclear cells as the most frequent aberrations.
